Take a direct regional or Intercity train from Roma Termini or Tiburtina, about 1 hour to 1 hour 20, then the funicular up to the town.
The funicular ride from Orvieto station is about two minutes and costs EUR 1.30. How do you get to Assisi by train?
Usually 2 to 2.5 hours from Rome with a change at Foligno; some direct trains run but are slower.
Assisi station is in Santa Maria degli Angeli, the lower town. From there a bus or taxi takes about 12 to 20 minutes up to the hill town and the Basilica of St Francis. Santa Maria degli Angeli, with the Porziuncola chapel, is near the station, so you can see it on the way in or out.

How long is the drive from Rome to Assisi?
About three hours each way by coach, according to the Greenline operator; Orvieto is about 1.5 hours from Rome on the way.
Times change with traffic, which is why the tours start early: 6:30 to 8:00 AM depending on the operator. One coach guest counted at least six hours on the bus and added that the description says so.
